What is Animal Models of Multiple Sclerosis about?

## Etiologies of MS and Its Animal Models Multiple sclerosis (MS) is an inflammatory demyelinating disease in the central nervous system (CNS). 1À3 Although the precise etiology of MS remains unclear, MS has been proposed to be a disease caused by interactions among autoimmunity, microbial infections, and/or genetic factors (Fig. 3.1). Clinically, the autoimmune etiology of MS has been supported by several findings: (1) anti-myelin T-cells and antibodies have been found in some MS patients 4 ; (2) T-cell and macrophage infiltration as well as antibody deposition are observed in demyelinating lesions 5,6 ; and (3) immunomodulatory and immunosuppressive drugs [disease-modifying drugs (DMDs)], such as interferon (IFN)-β, anti-very late antigen (VLA)-4 antibody, glatiramer acetate, anti-CD20 antibody, and fingolimod, are effective in some MS patients. 7,8 Experimentally, sensitization with CNS antigen can induce an autoimmune response in animals, resulting in MS-like CNS disease, experimental autoimmune encephalomyelitis (EAE). 9,10 Viral infections have also been associated with MS pathogenesis, 11,12 while other microbial agents, including microbiota, may play some roles. 13 Clinical
