What is iPSC-derived models of autism about?

Autism spectrum disorders (ASDs) are neurodevelopmental diseases caused by genetic and phenotypic heterogeneities that lead to deficits in verbal and nonverbal communication, impairment in social interaction, and the presence of restricted and stereotyped behaviors. Studies of syndromic autism are a valuable resource for understanding how a known genetic defect can be responsible for cellular and molecular alterations observed in ASD. Recently, induced pluripotent stem cell (iPSC)-derived models have enabled the generation of patient-derived neuronal tissue, allowing for the development of in vitro ASD cellular models. In this chapter, 2D and 3D iPSC-derived models from ASD patients will be presented, highlighting the findings obtained from syndromic autism studies. As brain organoids model early human brain development there is unprecedented potential to study several key aspects of brain formation, including neuronal differentiation, migration, neuronal activity, and interaction with cellular diversity.
