Experiments w i t h heterokaryons have demonstrated t h a t the d i f f e r e n t i a t e d state can be altered. Such short-term somatic c e l l hybrids stably r e t a i n all of the genes in their normal position w i t h i n chromosomes. Clsing this system, we have shown t h a t the expression of nine different muscle genes can be induced i n a range of human nonmuscle cells following fusion w i t h muscle cells. Differences i n t h e frequency and t i m e course of gene activation in endodermal, mesodermal and ectodermal c e l l types suggest differences in t h e lineage decisions and steps required t o generate and m a i n t a i n them. Diffusible molecules t h a t are transported t o nuclei through t h e cytoplasm presumably mediate t h e activation of genes observed in heterokaryons, since t h e nuclei of the t w o c e l l types r e m a i n separate and distinct. Changes in chromatin structure associated w i t h D N A replication do not appear necessary f o r --activation of muscle genes in all c e l l types tested, except H e L a cells.
