radiosensitivity was obtained in 83.8% (26/31) tumours. For 25 tumours, both assays were compared simultaneously and the success rates were 76% (19/25) with the Courtenay-Mills assay and 72% (18/25) with the ATCCS. The median SFz was 0.43 (range 0.16-0.77) with the Courtenay-Mills assay and 0.36 (range 0.22-0.61) with the ATCCS. There was, however, a poor correlation between the SF2 measurements obtained with these two methods r = 0.20). Fibroblast contamination was found to occur in both assays, and differences in the proportion of normal cells growing in the two assays may be one explanation for these differences. Results from repeat assays on HeLa cells suggest that inter-assay variability is another important contributing factor. Clonogenic assays are generally considered to be the standard method for measuring cellular radiosensitivity. In this study a high success rate was obtained with both assay methods but a wider range of values was found with the Courtenay-Mills assay than with the ATCCS.
