Wheat is a highly commercial and economically important crop that also serves as a major source of food for human consumption throughout the entire world. In North America, wheat farmers suffer major losses due to sawfly ( Cephus cintus) infestation, a pest that causes irreparable damage to wheat stems. Wheat varieties with solid stems have been found to be more resistant to sawfly, while their hollow-stemmed counterparts are highly susceptible to the pest. Consequently, stem solidity has become a trait of interest to wheat farmers, breeders and researchers. This is a comprehensive study of wheat stem solidity using histological and molecular biology techniques. The main objectives of this study were to characterize the solid stem trait and to identify genes that can be potentially used as markers for stem solidity. Select wheat cultivars varying in stem solidity, were morphologically examined by scanning electron microscopy along with a biochemical analysis of lignin content.
