Alkane 1-monooxygenase (alkB); n-Alkanes; Rhodococcus; Beta-galactosidase Rhodococcus sp. BCP1 grows on gaseous (C 2 -C 4 ), liquid (C 5 -C 16 ) and solid (C 17 -C 28 ) alkanes and it can also co-metabolize low-chlorinated solvents under aerobic conditions. A chromosomal region, including the alkB gene cluster, was cloned from Rhodococcus sp. BCP1 genome. The alkB gene cluster is homologue to the gene cluster family encoding an integral-membrane non-heme di-iron monooxygenase (alkB), two rubredoxins and rubredoxin reductase, acting as electron-transfer proteins. Retro-transcriptase-PCR analysis revealed that alkB gene cluster is harboured on a polycistronic operon whereas the transcription start of alkB was determined by primer extension procedure. In order to clarify the range of the substrates inducing the alkane hydroxylase expression, cells of Rhodococcus sp. BCP1 were transformed with a promoter probe vector containing the lacZ gene downstream of the alkB promoter. Resting cells of Rh. sp.
