The Enzymes of the Hepatic Phenylalanine Hydroxylating System by S. Kaufman is a Biochemistry, Genetics and Molecular Biology article available to read on EtoBox.
What is The Enzymes of the Hepatic Phenylalanine Hydroxylating System about?
The hepatic phenylalanine hydroxylating system has been shown to consist of at least three essential components, two enzymes-phenylalanine hydroxylase and dihydropteridine reductase--and a non-protein coenzyme, tetrahydrobiopterin (Kaufman, 1971). The reactions involved in the enzymatic conversion ofphenytalanine to tyrosine are shown in equations ( 1) and ( 2). Phenylalanine + O 2 + tetrahydrobiopterin --+ tyrosine + H20 + quinonoid dihydrobiopterin (1) Reaction ( 1) is catalysed by phenylalanine hydroxylase and reaction (2), which serves to regenerate the active, tetrahydro form of the coenzyme, is catatysed by dihydropteridine reductase (DHPR). Quinonoid dihydrobiopterin is an extremely unstable compound that undergoes tautomerization to 7,8dihydrobiopterin, an isomer, of the quinonoid compound (Kaufrnan, 1964). The 7,8-dihydrobiopterin is not a substrate fbr DHPR, but is a substrate for another enzyme, dihydrofolate reductase, which catalyses reaction (3) (Kaufman, 1967a).
