This document describes research on engineering Clostridium tyrobutyricum for n-butanol production. C. tyrobutyricum was engineered to overexpress aldehyde/alcohol dehydrogenase 2 (adhE2) from Clostridium acetobutylicum, which converts butyryl-CoA to butanol. Butanol titer of 1.1 g/L was achieved. The effects of inactivating acetate kinase (ack) and phosphotransbutyrylase (ptb) genes were then studied.
