Study hypothesis: Impaired brain water homeostasis contributes to brain edema in acute hepatic encephalopathy. Design: Prospective, controlled, randomized laboratory investigation. Methods: Fifty-one adult male Sprague-Dawley rats were randomized into experimental groups receiving daily IP injections of either 250 mg/kg thioacetamide (TAA, a liver toxin) or an equal volume of saline (control). Twentyfour hours after the second injection, half of the animals in each group received an IP injection of distilled water equal to 15% of their body weight. Animals were killed one hour later for determination of serum osmolality and ammonia; specific gravity; and cation contents and amino acid contents in cerebral gray matter, cerebral white matter, and basal ganglia. Data from animals receiving a water injection were compared (Student's t-test) with data from animals not receiving a water injection. Data were compared between animal groups (analysis of variance). Results: Serum ammonia in rats receiving TAA was twice that in control animals (P < .05).
