What is Hedgehog Signalling in Development and Disease about?
Hedgehog (HH) signalling is crucial for normal development and tissue homeostasis, and mutations affecting the pathway have been described in congenital malformations and cancers. Key steps in the pathway include ligand production and binding to its receptor, followed by the processing of the GLI transcription factors which feed into gene regulatory networks. Secreted over many cell diameters in some locations, HH ligands are classic morphogens, patterning tissues such as the neural tube and limb bud. Many small molecules have been shown to target the HH pathway, offering hope for the treatment of HH-associated disease. We extend this article by discussing dynamical properties of the HH signalling cascade. Objectives • To describe the basic structure of the hedgehog signaling pathway.• To discuss the role of cilia in hedgehog signalling.• To summarise evidence supporting non-canonical signalling pathways.• To introduce recent work modelling dynamical aspects of hedgehog signalling.• To discuss the genetic diseases caused by mutations in hedgehog pathway components.
