## Simple Summary Protein ubiquitination is catalyzed by many enzymes, whose functions and substrate specificity are not fully understood. This study reports the expression patterns of the membrane-associated RING-CH (MARCH) family members in breast cancer and their association with patient outcomes. Specifically, MARCH8 is a newly identified tumor suppressor with a role in inhibiting breast cancer metastasis and enhancing cancer cell death. MARCH8 not only promotes the degradation of membrane proteins such as the breast cancer stem-cell marker CD44 through the lysosomal degradation pathway, but also recruits a previously unknown nonmembrane target protein, signal transducer and transcription activator 3 (STAT3), for proteosome-dependent degradation. ## Abstract Protein stability is largely regulated by post-translational modifications, such as ubiquitination, which is mediated by ubiquitin-activating enzyme E1, ubiquitin-conjugating enzyme E2, and ubiquitin ligase E3 with substrate specificity.
