This study demonstrates that a stable leu2- yeast strain can be transformed to LEU2+ by a chimeric ColEl plasmid carrying the yeast leu2 gene. Experiments show that the ColEl and yeast DNA on the plasmid integrate into yeast chromosomes in multiple locations. This integration may be additive or substitutive, and the bacterial plasmid sequences then behave like a Mendelian element in yeast.
