What is A cellular platform enables targeted brain delivery of T cells about?
Miltenyi NK cell isolation kits. OS cells were treated with 10 ng/ml romidepsin (generously provided by Celgene) for 24 hrs. ALT-803 (3.5 ng/ml) was generously provided by Altor BioScience Corporation. NK proliferation, receptors expression and cytotoxicity were assessed (Chu/Cairo, et al., Can Imm Res 2015). 1 ug/ml or 10ug/ml of dinutuximab (generously provided by United Therapeutics) was used for ADCC assays. Results: Romidepsin significantly enhanced MICA/B expressions in SaoS-2, U2-OS, and HOS cells (p < 0.001). ALT-803 maintained the high expression of NKG2D of ExPBNK. Romidepsin, ALT-803 or in combination significantly enhanced exPBNK in vitro cytotoxicity in a E:T dependent manner (p < 0.001) against OS (Figure 1A). Romidepsin or ALT-803 significantly enhanced exPBNK mediated ADCC with dinutuximab in an E:T dependent manner (p < 0.001) against OS. The combination of romidepsin, ALT-803 and dinutuximab significantly enhanced exPBNK mediated ADCC and CD107a expression on exPBNK cells (Figure 1B) (p < 0.01) against OS. Conclusions: The combination of romidepsin and ALT-803 significantly enhanced exPBNK in-vitro cytotoxicity against OS. Romidepsin, ALT-803 or in combination sig
