This study identifies a complex structural variant associated with polled intersex syndrome (PIS) in goats, revealing a 10,159 bp deletion and a 480 kb duplicated segment affecting genes KCNJ15 and ERG. Long-read whole-genome sequencing enabled the development of a diagnostic PCR for genetic testing of polledness and PIS, confirming the variant in all 334 tested polled goats. The findings enhance understanding of the genetic basis of PIS and provide a reliable method for diagnosing this condition in goats.
