What is Histopaque-1077 for Lymphocyte Isolation about?
Histopaque-1077 is a sterile solution used to isolate mononuclear cells from whole blood through density gradient centrifugation. It contains polysucrose and sodium diatrizoate at a density of 1.077 g/mL. Whole blood is layered on top of Histopaque-1077 and centrifuged, which causes erythrocytes to pellet on the bottom while mononuclear cells remain at the interface. The interface is collected, washed, and resuspended for further use such as cell culture, HLA typing, or lymphocyte enumeration.
