This document describes a new method for measuring tissue sulfhydryl groups using a water-soluble derivative of bis(p-nitrophenyl) disulfide called bis(3-carboxy-4-nitrophenyl) disulfide (DTNB). DTNB reacts with sulfhydryl groups in a slightly alkaline medium to produce a highly colored p-nitrothiophenol anion that can be used to colorimetrically measure sulfhydryl concentration.
