An automated flow-injectron manifold is described for the kinetic determination of enzyme activities by a stopped-flow procedure with fluorescence detection. The linear calibration range for alkaline phosphatase is O-250 U 1-l with a precision of 2%; sample throughput is 35-40 h-'. Linear responses were also obtained for lipase (O-100 U l-'), acetylcholinesterase (O-500 U 1-l) and chymotrypsin (O-200 U 1-l). The advantages of this approach to the determination of plasma enzyme activities include sensitivity and the small sample and reagent volumes needed.
