## Historical perspective Antibody-drug conjugate (ADC) is one of the fastest-growing drug classes in oncology and a proven approach for treating cancer patients. Back in 1907, Paul Ehrlich, a German chemist and founder of chemotherapy, came up with the idea of Zauberkugel, which was translated to "magic bullet" in English [1]. With this idea, Ehrlich envisioned agents that go straight to their identified targets without harming the organism. More than a century later, antibody-based "magic bullets" are the most promising targeted agents that exist today. However, monoclonal antibody (mAb) alone demonstrates limited cytotoxic potency. To increase the cytotoxic power of these molecules, it was hypothesized that traditional chemotherapy and cytotoxic small molecules can be attached to mAbs to enhance their therapeutic effectiveness. These "enhanced" magic bullets will utilize the selectivity of mAb and possess potent cytotoxic small molecules that can kill the cancer cells. ADCs are essentially the realization of this concept and modern "magic bullets." ## Early development As shown in Fig. 7.1, researchers have been trying to make ADCs for over 100 years.
