Carbon dioxide (CO2) exists in the atmosphere and is formed by the respiration of all living organisms, fermentation of sugars, and burning of fossil fuels. An active area in synthetic chemistry is to capture this CO2 and use it to prepare high value-added fine chemicals, especially commercial drugs. Although the application of CO2 as a reagent in the synthesis is restricted owing to its kinetic inertness and chemical stability, numerous breakthroughs have been achieved in medicinal chemistry. This chapter summarizes the advances made in the past three decades in CO2-mediated carboxylation, methylation, and oxidation methodologies, in which CO2 is incorporated into C-, O-, and N-nucleophiles to form C–C, C–O, and C–N bonds, leading to the formation of active pharmaceutical ingredients and their key intermediates. This chapter provides a promising alternative to the known industrially driven methods and will be helpful in developing more promising CO2-based synthetic approaches.
