Prion disorders are a category of neurodegenerative illnesses caused by prions, which are misfolded proteins. Prions serve as a folding template for proteins during post-translational modification, transforming them into infectious prion form. Polymorphisms in the coding region of the prion protein gene influence susceptibility to some prion diseases, such as kuru or the new variant of Creutzfeldt-Jakob disease in humans and scrapie in sheep and goats, while other prion disorders, such as fatal familial insomnia disease in humans, have an underlying inherited genetic basis. PrPSc or misfiled form of the prion protein that accumulates in the central nervous system and, to a lesser degree, lymphoreticular tissues, is the sole biomarker for prion illnesses. There is no diagnostic test for prion diseases in live animals or people at this time. New diagnostic approaches are being developed to improve the sensitivity and specificity of PrPSc detection in bodily fluids and to find novel surrogate indicators.
