What is 086 Chd4 epigenetically regulates RORγt-dependent IL-17A production in γδT cells to promote psoriasiform skin inflammation about?
Type 2 cytokines, especially IL-4 and IL-13, play central roles in the pathogenesis of atopic dermatitis (AD). Immune cells in acquired immunity especially Th2 cells, and innate immune cells such as group 2 innate lymphoid cells (ILC2s) and basophils are the major source of type 2 cytokines in AD lesions. It is well known that alarmins including thymic stromal lymphopoietin (TSLP), IL-33, and IL-25 play essential roles in the activation of innate immune cells. However, it is not clear whether alarmins also play roles in the activation of Th2 cells in AD lesions. To investigate the involvement of alarmins in the activation of Th2 cells in AD lesions, we collected CRTH2 + Th2 cells in the skin biopsy samples from 11 AD patients. After expanding T cells in vitro using IL-2 for 4 days, we stimulated T cells with a cocktail of several alarmins (TSLP, IL-33, IL-25, IL-1b, IL-18) and/or TCR stimulation by anti-CD3/CD28 for 24 hours. Then we performed a flow cytometric analysis of cytokine production (IL-4, IL-13) from CRTH2 + Th2 cells in each group. We found that the stimulation with the alarmin cocktail alone did not induce cytokine production from CRTH2 + Th2 cells. However, stimulatio
