Phosphate buffers consist of mixtures of monobasic dihydrogen phosphate and dibasic monohydrogen phosphate salts that can buffer between pH 5.8-8.0. Phosphate buffers have a high buffering capacity but can inhibit enzymatic reactions, prevent DNA/RNA precipitation with ethanol, and sequester divalent cations. To prepare a 0.1M potassium phosphate buffer at 25°C, specific amounts of K2HPO4 and KH2PO4 salts are mixed with water.
