This document describes a new chemical reaction that allows the synthesis of novel tubulysin analogues with enhanced cytotoxicity. The key step uses dibutyltin oxide catalyst to transform an N,O-acetal ester to an N,O-acetal ether and facilitate hydrolysis of a C-terminal ester. This enables an expedient synthesis of new tubulysin analogues and their activated hydrazides.
