This study compares the 3,5-dinitrosalicylic acid (DNS) reducing sugar method and the Nelson-Somogyi (NS) method for measuring endo-1,4-β-xylanase activity using various xylan substrates. The authors found that the DNS method significantly overestimates enzyme activity due to secondary reactions, while the NS method accurately reflects the quantity of glycosidic bonds cleaved. They recommend discontinuing the use of the DNS method in favor of the more reliable NS method for endo-xylanase measurement.
