What is Characterization and Kinetic Mechanism of Mono- and Bifunctional Ornithine Acetyltransferases from Thermophilic Microorganisms about?
The __argJ__ gene coding for __N__^2^‐acetyl‐l‐ornithine: l‐glutamate __N__‐acetyltransferase, the key enzyme involved in the acetyl cycle of l‐arginine biosynthesis, has been cloned from thermophilic procaryotes: the archaeon __Methanoccocus jannaschii__, and the bacteria __Thermotoga neapolitana__ and __Bacillus stearothermophilus__. Archaeal __argJ__ only complements an __Escherichia coli argE__ mutant (deficient in acetylornithinase, which catalyzes the fifth step in the linear biosynthetic pathway), whereas bacterial genes additionally complement an __argA__ mutant (deficient in __N__‐acetylglutamate synthetase, the first enzyme of the pathway). In keeping with these __in vivo__ data the purified His‐tagged ArgJ enzyme of __M. jannaschii__ only catalyzes __N__^2^‐acetylornithine conversion to ornithine, whereas __T. neapolitana__ and __B. stearothermophilus__ ArgJ also catalyze the conversion of glutamate to __N__‐acetylglutamate using acetylCoA as the acetyl donor. __M. jannaschii__ ArgJ is therefore a monofunctional enzyme, whereas __T. neapolitana__ and __B. stearothermophilus__ encoded ArgJ are bifunctional. Kinetic data demonstrate that in all three thermophilic organisms
