To de&mine the antigeniclty of the cell binding site within a formal toxoid, the competition of the toxoid for the neutralizing antibodies which prevent cell-toxin association was investigated in a cell culture assay. In the assay developed we measured the fraction of the antibody bound site released by a toxoid added to a mixture containing toxin and anti-toxin. The measured fraction is a function (E) which expresses the quantity and quality of both the antitoxic antisera and the toxoid. In defined systems where the only variable is the toxoid. the function (E) becomes a function in the concentration and affinity of the toxin-binding site structures within the toxoid, which we termed the competition index (Q). Investigations on four toxoids revealed a correlation between the toxoid competition index (Q), the toxoid in viw potency, and the neutralization titers of the antibodies developed by the toxoid to the toxin binding site. The results demonstrate that the competition index (Q) is a reliable parameter are needed for formulation of the exact correlation and the identification of a competition index value to certificate a vaccine.
