It is an attempt at summarizing the data available on synthetic antigens as well as known synthetic adjuvants (including possible modes of action) and focuses on a potential new class of adjuvants, namely polycationic peptides. Different approaches toward better-defined vaccines, such as subunit vaccines, which use purified or recombinant proteins derived from pathogens, and DNA vaccines have been reviewed recently and are not the topic of this review article (Ben-Yedida and Arnon, 1997;Donnelly et al., 1997). Well-defined, but non-synthetic adjuvants, such as conjugates of cholera toxin (from Vibrio cholerae) and heat labile enterotoxin (from Escherichia coli), interleukins, immune stimulatory complexes (ISCOM) and compounds purified from natural sources (saponins, QS 21, monophosphoryl lipid A), will not be covered either.
