The monoclonal antibodies 66C7 and 88E8, both specific for human salivary <x.-amylase, inhibit by their combined synergistic action human salivary ,-amylase by more than 97%. Pancreatic OL-amylase is not influenced by the two antibodies. Two reagents using the two antibodies for pancreatic oL-amylase determination are described. The Substrates for the activity determination are p-nitrophenyl-maltoheptaoside and the ethylidene protected analogue. The direct determination of human pancreatic ot-amylase without calculation from total and residual activity after partial Inhibition or by use of various Separation Steps allows that the parameter pancreas a-amylase will in future replace the total ÖL-amyläse determination. This is a further Step in the differential diagnosis of pancreatic diseases.
