One of the foremost problems challenging biochemists today is the understanding of integrated metabolic systems as functional units within the cell, & the factors which influence & control them. The major pathways of metabolism in such systems have been established over the past thirty years, & the University of Pennsylvania has been fortunate to be a contributor to many important aspects through some of its distinguished faculty, such as Otto Meyerhof, W. C. Stadie, & D. Wright Wilson. It is fitting that an evaluation of the present status of the field be made during this 200th anniversary of the founding of the School of Medicine at the University of Pennsylvania. Present knowledge of the dynamics of cellular metabolism is based largely upon an extension of the basic principles of physical chemistry as applied to enzyme reactions in solution. The establishment of current hypotheses spans the metamorphosis from classic studies with single enzymes to the understanding of the mechanism of integrated enzyme reactions.
