Suitability of the orthophosphate determination method developed by Murphy and Riley ((1963) Am/. Chin!. Acttr 27, 31-36) for ATPase assays was investigated. This method was simple and sensitive as compared with the usual other methods. However, ATP. EDTA. EGTA. KCI. reducing agents. mannitol, sorbitol. and Triton X-100 interfered with the color development. Thus. for the estimation of orthophosphate in the presence of interfering chemicals. the concentration of the latter must he kept at below inhibitory levels.
