This study developed a new method to directly quantify discrete plasmid transfer events between bacteria by using large numbers of replicate matings in microtiter wells and counting the number of transfer-positive and transfer-negative wells. The method was used to study the effects of mercury and 2,4-D on the transfer of the IncP1 plasmid pRO103 from Escherichia coli to Pseudomonas putida, finding that mercury above 1.0 mg/L negatively affected transfer by stressing the recipient.
