with late toxicity in patients with prostate cancer who received external beam radiotherapy. They observed a greater prevalence of late Grade 2 or higher rectal and/or genitourinary side effects in carriers of the VEGF À634 G/ C SNP than in noncarriers in their population of men treated with external beam irradiation for prostate cancer. There is overlap in our studies that is worth noting. There is strong evidence that transforming growth factor b (TGF-b) and hypoxia physically interact to induce transcription of VEGF (1). The TGF-b acts through Smad proteins as intracellular signaling effectors (2). The TGF-b stimulates VEGF in patients with certain malignancies to enhance a tumor's malignant potential (3, 4). As described in both our study and the work by Langsenlehner et al., it also appears that both TGF-b and VEGF are implicated in the development of late effects after radiation therapy. One can hypothesize that certain polymorphisms in TGFB1 mediate late effects after radiotherapy because of its profibrotic effects, and certain SNPs in VEGF lead to increased late effects related to angiogenesis. We strongly believe the next logical and useful step is the performance of genom
