What is TGF-β Family Signaling about?

Members of the transforming growth factor b (TGF-b) family of growth and differentiation factors play critical roles in embryonic development and adult tissue homeostasis, and lead to a number of serious human diseases when their downstream signaling is misregulated. In this review we discuss the main components of the TGF-b family signaling pathways and their regulation. Starting with a discussion of the production and release of ligands, we go on to outline their binding to and activation of cell surface receptors and the subsequent signal transduction to the nucleus via the SMAD proteins. We end with a discussion of the role of deregulated TGF-b family signaling in human disease.
