healing response stimulates proliferation. Initial tissue damage by radiation is generated by direct reactive oxygen species (ROS) action on DNA as well as by initiation of cell signaling pathways. The cell signaling pathways may initiate the wound healing response, characterized by an increase in permeability, edema and fibrin accumulation in extracellular matrix. This is followed by macrophage infiltration, accumulation and activation. Focal hypoxia in regions of macrophage accumulation, suggest that the mechanism responsible is high oxygen consumption, as would occur with creation of ROS. ROS and hypoxia are likely to further stimulate the profibrogenic and proangiogenic pathways. Hypoxia therefore continuously perpetuates non-healing tissue response leading consequently to chronic radiation injury. Targeting ROS in the initial phase of injury (as we have done using strong antioxidant MnTE-2PyP) or during the hypoxia ROS generated period might be an effective way to modulate this response.
