What is Analytical evaluation of the Abbott ARCHITECT Sirolimus assay about?

Comparison against immunoassay (x) without and with urine extraction gave the following regression equations, respectively: LC-MS/MS = 0.24x -54 and LC-MS/MS = 0.72x -3. Concentrations of urinary free cortisol by LC-MS/MS were much lower than immunoassay (Advia Centaur) without urine extraction (10% of immunoassay values). When compared to immunoassay (Immulite 2000) with urine extraction the results were closer in value (67% of immunoassay values). Close correlation was found with an external lab's LC-MS/MS method. Normal individual results fell within reported literature reference ranges. Conclusion: LC-MS/MS is a more sensitive and specific method with a wider reportable range for urinary cortisol as compared with immunoassay. If immunoassay is to be used for urine free cortisol, prior solvent extraction is required.
