This document describes a new method for extracting ancient DNA from low-quality samples that improves DNA yields while increasing sample throughput. The method combines the high DNA yields of batch-based silica extraction with the time efficiency of column-based methods. Testing on Pleistocene cave bear samples showed DNA yields were comparable to silica batch extraction but processing multiple samples in parallel decreases time and costs.
