What is SUPPLEMENTAL MATERIAL Supplemental Methods for Dusp14-Mediated Dephosphorylation of MLKL about?

In this study, male wild-type C57BL/6 mice were sourced from the Peking University Medical School Experimental Animal Center. This study was approved by Peking University Institutional Animal Care and Use Committee (PURB-LA2023086). Dusp14 knockout mice with a C57BL/6 background were obtained from Cyagen Biosciences Inc (Suzhou, China) and were further bred by our research group. All mice were housed at the Peking University Medical School Experimental Animal Center under controlled environmental conditions, which included a temperature of 23°C humidity maintained at 50-60%, and a 12-hour light-dark cycle. For the isolation of neonatal mouse primary cardiomyocytes, C57BL/6 neonatal mice aged 1-3 days were acquired from the Peking University Medical School Experimental Animal Center. To induce hypothyroidism, 6-propyl-2-thiouracil (PTU, Sigma #P3755) was dissolved in a sodium hydroxide solution (1 mol/l) and subsequently added to the drinking water of the hypothyroidism (HT) group. The pH of the water was adjusted to maintain a range of 7-8 using hydrochloric acid titration, and the final PTU concentration in the drinking water was set at 0.025%. To prevent PTU precipitation and ensu
