PARP inhibitors show promise for treating cancers with defective DNA repair pathways like BRCA mutations. In clinical trials, the PARP inhibitor olaparib demonstrated a 46% response rate in BRCA-mutated ovarian cancer with a median response duration of 8 months and was well tolerated. Further studies are exploring olaparib as a maintenance therapy and in combination with chemotherapy. Biomarkers are needed to identify which sporadic ovarian cancers may also benefit from PARP inhibition due to "BRCAness".
