Elevation high plasma cholesterol plays a significant role in promoting the incidence of atherosclerosis and coronary heart disease. In the current study, a high cholesterol diet (HCD) rat model for cholesterol homeostasis investigated the effect of pigeon pea by-product protein hydrolysate (PPH) and derived potential peptide (PFVKSEPIPETNNE) (PEP). Potential evidence for cholesterol homeostasis was produced by the lipid profile, mRNA expression, protein expression and histopathology results. The transcription of the 3-hydroxy-3-methylglutaryl coenzyme A reductase enzyme (HMGCR) and low density lipoprotein receptor (LDL-R) that absorbs cholesterol through receptor-mediated endocytosis is regulated by the sterol regulatory element-binding protein (SREBP-2) isoform. These studies show that PPH has greater potential to restore transcriptional and post-transcriptional changes in regulatory markers by rat administration compared to PEP, and provide a rational explanation for the effect of PPH on the treatment of hypercholesterolemia.
