The search for molecules involved in T-cell-mediated cytotoxicity is increasingly making use of molecular biology approaches. One of these approaches is to try to identify molecules specifically present in cytolytic T cells (Tc) and not in other cells. In theoretical terms, no initial assumption is thus made as to which molecules may be at play, other than tissue-specificity. In practical terms, the search tor such molecules takes the form of an identification of genes with tissue-restricted transcription, with the secondary need of establishing relationship to function. ## Methods The general principle is to identify mRNA transcripts ideally present in cytolytic T cells and not in other cells. In practice (Table 1), cDNA libraries (either sub tracted or not) are subjected to differential screening (with radioactive cDNA probes, either from Tc or from other cells, or subtracted). The minority Tc + non-Tc clones are grouped according to cross-hybridization patterns and checked for tissue-distribution usually with Northern blots.
