In this study, two series of experiments were carried out to elucidate the mechanism of this adverse effect: high-speed observation of deformation of microbubbles and cells exposed to ultrasound and evaluation of the resulting cell damage using fluorescence microscopy. Methods: In one series of experiments, microbubbles and endothelial cells were exposed to pulsed ultrasound of 1 MHz in center frequency and 0.6 MPa in peak rarefactional pressure and observed as 24-frame sequential images at a frame rate of 4 million fps. In another series of experiments, endothelial cells were exposed to ultrasound pulse with microbubbles, and the damage to the cells was evaluated by fluorescent staining using propidium iodide. Plastic-shelled microbubbles and Levovist® were used in the experiments. Results: In the high-speed images, generation of a small stream of surrounding liquid caused by nonuniform contraction of a microbubble and deformation of a cell beside the bubble were observed.
