What is PVN-NAc shell-VP circuit OT and OTR neurons regulate pair bonding via D2R and D1R about?
Abstract34 Previous studies have found that several neurochemicals are involved in 35 formation of pair bonding. However, circuit mechanisms underlying pair bonding, 36 especially how these chemicals interact in this circuit regulate pair bonding, remains 37 unclear. Using male mandarin voles, the present study shows that cohabitation with a JNeurosci Accepted Manuscriptpartner increased frequency of spontaneous excitatory postsynaptic current (sEPSC)38 of paraventricular nucleus (PVN) oxytocin (OT) neurons projecting to nucleus 39 accumbens (NAc) shell. Optogenetic activation of PVN OT neurons projecting to the40 NAc shell reduced D2 medium spiny neurons (MSNs) activity, but increased activities 41 of D1 MSNs in the NAc shell. Bath application of OT caused a long-term depression 42 (LTD) of evoked excitatory postsynaptic current (eEPSC) in NAc shell D1/D2 MSNs 43 in the no cohabitated male voles. This OT-induced LTD in the NAc shell D1/D2 44 MSNs was suppressed by 7 days of cohabitation. NAc shell oxytocin receptor (OTR)45 MSNs projecting to the ventral pallidum (VP) were D1R/D2R positive. Chemogenetic 46 activation or inhibition of OTR MSNs in the NAc shell projecting to the VP f
