This study investigates the role of the PI3K/AKT/mTOR signaling pathway in the differentiation of myofibroblasts from valvular interstitial cells in canine myxomatous mitral valve disease (MMVD). It demonstrates that TGF-β1 induces this transition through the upregulation of the PI3K/AKT/mTOR pathway, which is associated with cellular senescence and altered secretory phenotype. The findings suggest that targeting this pathway may offer new therapeutic strategies for managing MMVD in both dogs and humans.
