1) E. coli was genetically engineered to produce the sialic acid precursor CMP-Neu5Ac from its own metabolism instead of relying on exogenous sialic acid. Mutant strains lacking Neu5Ac aldolase and ManNAc kinase were able to efficiently produce 3-sialyllactose. 2) A concentration of 25 g/L of sialyllactose was obtained in high cell density fermentation with continuous lactose feeding. 3) This reduces production costs and makes sialylated oligosaccharides feasible for applications like the food industry.
