The study investigates the role of cathepsin B in the efficacy of antibody-drug conjugates (ADCs) with a protease-cleavable valine-citrulline (VC(S)) linker. Contrary to expectations, the suppression of cathepsin B did not impact ADC effectiveness, suggesting that other cysteine proteases can also cleave the VC(S) linker and that ADC efficacy is more dependent on the antibody and intracellular targets.
