What is D69. ASTHMA SUBPHENOTYPES, TARGETS AND THERAPEUTICS - TRPM8 Expression in the Airway Epithelium Is Related to Cold-Induced Airway Hyperresponsiveness and Mucus Hypersecretion in Asthma about?
RATIONALE. Cold temperature is an important environmental factor influencing airway inflammation and lung function in patients with asthma and COPD. Previous studies established possible molecular pathway for TRPM8-mediated cold-induced mucus hypersecretion but clinical data is currently limited. The aim of this study was to assess the expression of TRPM8 mRNA in the nasal epithelium of asthma patients in connection with cold-induced airway hyperresponsiveness and expression of mucins MUC5AC and MUC5B. METHODS. The study enrolled 55 patients including 37 (67%) with mostly uncontrolled mild-to-moderate asthma and 18 (33%) with chronic bronchitis. Mean age of the patients was 38.9±1.69 years, 65% were females. Nasal epithelium specimens were collected by brush-biopsy and stored in RL-buffer at -80°C until RNA extraction. Expression of TRPM8, MUC5AC and MUC5B was evaluated by RT-qPCR before and after the bronchoprovocational challenge with 3-min isocapnic (5% CO2) cold air (-20°C) hyperventilation (ICAH) through the nasal mask. Cold-induced airway hyperresponsiveness was diagnosed by ICAH performed through the mouthpiece; FEV1 fall by 10% or more from the baseline was considered as a
