Releasing oligosaccharides from glycoproteins by hydrazionolysis is a reliable method for quantifying N-linked oligosaccharides . Compared with N-glycosidase digestion, hydrazionolysis is advantageous in obtaining high levels of recovery regardless of the protein's stereo conformation, but this goes hand in hand with the disadvantage of losing N-acetyl moieties of glucosamine or neuraminic acid during the reaction. These damaged sugars must be recovered before oligosaccharides are labeled by Xuorescent derivatives because they cannot withstand the severe conditions of the labeling reaction. The conditions of the acetylation reaction must be kept at weak alkali, but the pH balance of the solution decreases over the course of the reaction due to the acetic acid released from acetic anhydride. To prevent a decrease in the pH level, excess salt (e.g., sodium hydrogen carbonate) is normally added to the solution. However, removing the salt after the reaction is so laborious that this process needed to be improved to increase performance.
