Impregnation, also called infiltration, is the process of removing the clearing agent from tissue and replacing it with paraffin wax or another medium to firm the tissue for sectioning. Paraffin wax is most commonly used as it allows thin sections to be cut without distortion and tissues can be stored indefinitely. Tissues are processed manually or automatically through steps of dehydration, clearing, and infiltration to fully replace tissue fluid with wax. Vacuum infiltration can speed the process.
