This study explores the use of CRISPR-Cas9 for selectively targeting somatic mutations in pancreatic cancers, focusing on mutations that create protospacer adjacent motifs (PAMs). Whole genome sequencing revealed an average of 417 somatic PAMs per tumor, leading to significant selective cell death in pancreatic cancer cell lines without off-target effects. The findings suggest CRISPR-Cas9 as a promising strategy for cancer-specific therapies by exploiting noncoding somatic mutations as therapeutic targets.
