What is Discovery of microRNA–mRNA Interaction Through Genomics and Computational Prediction about?

microRNAs play essential roles in RNA silencing and regulating gene expression, and their involvement has been demonstrated in normal and pathological processes. microRNAs function by binding to the target mRNA molecule. The dysfunction of microRNAs has been associated with the development and progression of many diseases. To better understand the function of microRNAs in biological processes, we have to identify the interacting targets for the microRNAs. As there are millions of interactions between microRNAs and mRNAs, simple experimental approaches to identify microRNA–mRNA interactions would be extremely expensive and even infeasible. Therefore, computational prediction of microRNA targets is critical in identifying candidate miRNA–mRNA interactions for further experimental validation. The computational microRNA target prediction tools encompass a range of approaches. Some methods are based on canonical features while some other methods utilize emerging new microRNA binding experiments to discover novel features. In this chapter, we will review major methods in each of the categories and provide a comprehensive assessment of those tools. Understanding the basis of those microRN
